ABSTRACT

Chitosanases represent a class of glycoside hydrolases with high catalytic activity on chitosan but nearly no activity on chitin. Chitosanases can convert high molecular weight chitosan into functional chitooligosaccharides with low molecular weight. In recent years, remarkable progress has been made in the research on chitosanases. This review summarizes and discusses its biochemical properties, crystal structures, catalytic mechanisms, and protein engineering, highlighting the preparation of pure chitooligosaccharides by enzymatic hydrolysis. This review may advance the understandings on the mechanism of chitosanases and promote its industrial applications.

ABSTRACT

Obejctive To determine and compare britanin content in dried aerial parts and capitulum of Inula linariifolia Turcz from 7 different origins by HPLC. Methods Analysis was performed on Agilent Zorbax SB C18 (250 mm×4.6 mm, 5 μm). Acetonitrile and water were used as mobile phase for gradient elution at 1.2 ml/min. Column temperature was 30 ℃ and the detection wavelength at 212 nm. Results The results meet the requirements of the method validation in 2015 edition of Chinese Pharmacopeia. The average britanin content in dried aerial parts of Inula linariifolia Turcz is 0.125% vs 0.732% in capitulum, which is significantly different. Conclusion The established method is simple and convenient. It can be used for quality control of Inula linariifolia Turcz.

ABSTRACT

OBJECTIVE@#To construct a HIV-1 gp120 transgenic mouse model (gp120) with 7 nicotinic acetylcholine receptor (7nAChR) gene knockout.@*METHODS@#The 7nAChR gene knockout mice (7R) were crossed with HIV-1gp120 transgenic mice (gp120) to generate F1 generation mice. We selected the F1 mice with the genotype of 7R/gp120 to mate to obtain the F2 mice. The genotypes of the F3 mice were identified by PCR, and the protein expressions in the double transgenic animal model was analyzed by immunohistochemistry. BV2 cells were treated with gp120 protein and 7nAChR inhibitor, and the expressions of IL-1β and TNF- were detected using ELISA.@*RESULTS@#The results of PCR showed the bands of the expected size in F3 mice. Two F3 mice with successful double gene editing (7R/gp120) were obtained, and immunohistochemistry showed that the brain tissue of the mice did not express 7 nAChR but with high gp120 protein expression. In the cell experiment, treatment with gp120 promoted the secretion of IL-1β and TNF- in BV2 cells, while inhibition of 7nAChR significantly decreased the expression of IL-1β and TNF- ( < 0.001).@*CONCLUSIONS@#By mating gp120 Tg mice with 7R mice, we obtained gp120 transgenic mice with 7nAChR gene deletion, which serve as a new animal model for exploring the role of 7nAChR in gp120-induced neurotoxicity.

ABSTRACT

Objective To analyze the changes of radioactivity of the marginal zone of hepatic alveolar echinococcosis (HAE) before and after the treatment with albendazole liposomes,and to explore the value of 18F-fluorodeoxyglucose (FDG) PET/CT imaging in therapeutic response.Methods From July 2011 to July 2015,37 patients (21 males,16 females;average age (53±12) years) with HAE were selected and underwent 18F-FDG PET/CT imaging before routine medication with albendazole liposomes (10 mg · d-1 · kg-1),and the maximum standardized uptake value (SUVmax) of the margin of lesions was calculated.PET/CT imaging was performed again 6 months later to measure the SUVmax of the marginal lesions.Two-sample t test was used to analyze the data.Results In 37 patients with HAE,SUVmax before treatment was 4.88± 1.86,which was higher than that of 24 patients with reduced lesion size after treatment (3.63± 1.38;t =5.24,P< 0.001).Conclusion 18F-FDG PET/CT is useful for evaluating the radioactivity of marginal zone of HAE before and after the treatment with albendazole liposomes,and SUVmax of the marginal zone could be valuable for therapeutic response evaluation.

ABSTRACT

[Objective]To observe the dynamic changes of serum LH levels in different GnRH-along protocols ,and investigate the relationship between GnRH-adosage and LH levels ,and compare the clinical outcomes among different GnRH-a long protocols.[Methods]In this retrospective study,1.0 mg,0.8 mg,0.375 mg long-acting and 0.1 mg/d,0.05 mg/d short-acting GnRH-a long protocols were included from January to June in 2015 at the Reproductive Medicine Center of the First Affiliated Hospital of Sun Yat-Sen University. Serum LH levels were evaluated from a total of 250 women at the four time points,on the day before gonadotropin stimulation(Gn0),the fourth day of Gn stimulation(Gn4),the seventh day of Gn stimulation(Gn7),and the HCG administration day(HCG day),then the relationship between serum LH levels and the dosage of GnRH-a were analyzed. The number of oocytes retrieved,fertilization rate,good quality embryos rate,blastocyst transferred rate,the number of transferred embryos,implantation rate and clinical pregnancy rate were also compared.[Results]Among the long-acting groups,LH levels in 0.375 mg group were higher than those in 1.0 mg and 0.8 mg groups at the four time points(P0.05). In short-acting groups,the LH levels in 0.05 mg/d protocol were significantly lower than those in the 0.1 mg/d group at Gn0,Gn7 and HCG day(P0.05).[Conclusion]Among the long-acting groups,the smaller amount of GnRH-a was administrated ,the higher LH levels during ovarian stimulation and implantation rate the patients ob?tained. As to the short-acting groups,the LH level and implantation rate in 0.1 mg/d group is higher than those in 0.05 mg/d group.

ABSTRACT

BACKGROUND:As the bone and marrow tissue have very special structure, it is difficult to simultaneously display the bone with tough hard tissue and bone marrow tissues containing various immature hematopoietic cel s in the conventional process of pathological section preparation. OBJECTIVE:To choose the best decalcifying solution that cannot only completely remove the calcium in the bone tissue but also protect the structure of bone marrow tissues and cel s from damage. METHODS:Bone marrow tissues from the long bone of dogs were randomly divided into four groups. Under the same conditions, the bone marrow tissues were decalcified with 14%formaldehyde saline solution of nitric acid (group A), 14%nitric acid solution (group B), 20%A saline solution of hydrochloric acid formaldehyde (group C) and 20%A hydrochloric acid aqueous solution (group D). Decalcified time was recorded, fol owed by routine dehydration, section, hematoxylin-eosin staining and microscopic observation. Pathological section quality and hematoxylin-eosin staining were compared among the four groups. RESULTS AND CONCLUSION:Group A had the best sections and hematoxylin-eosin staining, strongest decalcified ability, shortest decalcified time and minimum damage to the bone marrow. Group B had the worst results of section and hematoxylin-eosin staining, in which, the bone tissues were loose and became yel ow and the bone marrow tissue were damaged greatly, and the decalcified effect was worse. Group C was worse than group A in decalcified ability, damage degree, section quality and hematoxylin-eosin staining results. Group D also had a better result of section and hematoxylin-eosin staining as wel as exhibited uniform decalcification effect and less damage to the bone marrow, which was ranked between group B and group C. Al the four kinds of decalcifying solutions have a good decalcification ability, but the section quality and hematoxylin-eosin staining results rank as fol ows:Group A>Group C>Group D>Group B. Taken together, 14%formaldehyde saline solution of nitric acid is ideal for the clinical preparation of pathological sections.

ABSTRACT

BACKGROUND:Breviscapine has been shown to impact the reproductive capacity in rats with type 2 diabetes mel itus, but few reports concerned its mechanism of action. OBJECTIVE:To study the effects of breviscapine on proliferating cel nuclear antigen and proto-oncogene c-fos expression in testis of type 2 diabetes mel itus rats. METHODS:Total y 36 healthy male rats were randomly divided into control group, model group and breviscapine group with 12 rats in each group. In the model group and breviscapine group, rat models of type 2 diabetes mel itus were established by continuous intraperitoneal injection of streptozotocin. Blood glucose reaching 16.7 mmol/L in rats was considered as the standard of model induction. In the control group, rats were given an equal volume of citrate buffer solution by single intraperitoneal injection. In the breviscapine group, rats were administered breviscapine 10 mg/(kg?d) for 4 consecutive weeks by intraperitoneal injection. Rats in the other two groups were injected with an equal volume of physiological saline at the same time point. RESULTS AND CONCLUSION:After 4 weeks of intervention, serum testosterone testing, immunohistochemistry and PCR results showed that serum testosterone levels, proliferating cel nuclear antigen, c-Fos protein and mRNA expression:control group>breviscapine group>model group (P<0.05);blood glucose concentration:the control group

ABSTRACT

Objective To investigate the clinical characteristics and surgical treatment of primary bone non-Hodgkin′s lymphoma.Methods Recalling a case of primary bone non -Hodgkin′s lymphoma patient data in Affiliated Maojian Hospital of Hubei University of Medicine .This patient was hospitalized because of pain in the right upper arm.By improving the relevant examination and diagnosis of right humeral bone tumors ,We had successfully completed a bone tumor resection and graft as well as dual plate fixation .Results Pathological diag-nosis after surgery showed the primary non -Hodgkin′s lymphoma of bone ,9 months follow-up after surgery ,the treatment effecincy was good .There were no tumor recurrence and distant metastasis .Conclusion Primary non-Hodgkin′s lymphoma of bone is uncommon in clinical practice .If there is primary bone tumor with no obvious sys-temic symptoms in elderly patients , we should considered it as a possible diagnosis of primary bone non -Hodgkin′s lymphoma,in order to reduce the possibility of misdiagnosis and mistreatment .Short-term effects of surgical treatment of this disease is excellent .

ABSTRACT

The in-vitro fertilization and embryo transfer technique has been widely applied in human insemination. The rate of successful insemination is gradual y rising, and the in-vitro fertilization directly determine the insemination outcome. OBJECTIVE:To evaluate the difference between the two common using insemination methods, microdrop and open, in in-vitro fertilization and embryo development. METHODS:A randomized study was conducted to compare microdrop and open insemination methods among non-male factor patients undergoing in-vitro fertilization and embryo transfer. A total of 1 175 cases were enrol ed in the research. There were 573 cases in the microdrop group, and 602 cases in open insemination group. The fertilization rate and embryo development in the two groups were compared. RESULTS AND CONCLUSION:The fertilization failure rate [total fertilization failure rate+low fertilization rate (0.05). The open insemination method is a simple insemination method with a lower fertilization failure rate. As the fertilization is a highly complicated process involving many extrinsic and intrinsic factors, further study is needed to confirm the effects of the two insemination methods on in-vitro fertilization outcome.

ABSTRACT

Objective To find out the risk factors of postoperative low cardiac output syndrome(LCOS) of patients undergoing cardiac valvular surgery in ICU in order to provide basic for prevention and control measures.Methods Ninety-six valve replacement patients with valvular heart disease were enrolled as our subjects and they were hospitalized in ICU of the First People's hospital of Yichang from Jan.2008 to May.2013.The patients postoperative LCOS (Dopamine > 10 μg/(kg · min)) were served as observation group (n =41),and the other were control groups(n =55).All data of the patients were recorded.Non-conditions Logistic regressions analysis were adopted to analyze the independent risk factors which resulted in LCOS undergoing cardiac valvular surgery.Results Of 96 patients undergoing cardiac vavular surgery,41cases (42.7%) had postoperative LCOS.Single factor analysis showed that hepatomegaly (P =0.007),course of diseases ≥ 15 years (P =0.042),cardiopulmonary bypass ≥ 120 min (x2 =3.937,P =0.047),pre-operative cardiac function ≥ Ⅲ degree (P =0.003) were the independent risk factors of postoperative LCOS undergoing cardiac valvular surgery.The Logistic multi factor regression analysis showed that the independent risk factors of postoperative LCOS undergoing cardiac valvular surgery included course of diseases ≥ 15 years (OR =2.825,95% CI =(1.015-7.861)),Pre-operative cardiac function ≥ Ⅲ degree (OR =7.306,95% CI =(2.050-26.035),P=0.002).Conclusion Course of diseases ≥15 years and Pre-operative cardiac function≥ Ⅲ-Ⅳ degree are the independent risk factors of LCOS undergoing cardiac valvular surgery.

ABSTRACT

Objective To explore the current status of humanistic care among medical students in clinical practice in obstetrics and gynecology department and its related influencing factors and to propose the corresponding countermeasures.Methods Scale of humanistic care quality was used in the survey for clinical practice students in obstetrics and gynecology department.ResultsThe total score rate of human care quality of medical students before clinical teaching were (84.12 ± 9.24)with a scoring rate of 72%.Significant differences were observed in the medical students with different medical professional(P <0.05).The total score of humanistic care quality of medical students after clinical teaching were (96.41 ± 1 1.53 ),significant higher than that of before training ( P < 0.05 ).ConclusionHumanistic care quality in medical students needs to be strengthen.Clinical training in obstetrics and gynecology department combined with humanities education has important practical significance to improve the quality of medical students.

ABSTRACT

Objective To analyze the curative effect of adult urethrocutaneous fistula after hypospadias repair with X-V dartos flap technique.Methods In the experimental group,the closed fistulas were wrapped with X-V dartos flap in the 22 cases of adult hypospedias fistulas from February 2007 to October 2010.In the control group,the routine fistulas repairs were preformed in the 35 cases of adult hypospadias fistulas from January 2000 to January 2007.The operative effects were compared between the two groups.Results All cases were followed up for 3-49(5.6±12.0)months,no fistula was found in the experimental group,while 7 fistulas(20.0%,7/35)were found in the control group.The fistulas rate had statistic difference between the two groupe(P<0.05).Conclusion The use of the X-V dartos flap technique can prevent adult urethroeutaneous fistula after failed hypospadias fistulas repair and raise the success rate.

ABSTRACT

Objective To investigate the effect of hyperthermia and mild hypothermia on permeability of blood-brain barrier(BBB),activity of matrix metalloproteinase-9(MMP-9)and expression of laminin in rats with severe traumatic brain injury(TBI)so as to explore the mechanism of hyperthermia and mild hypothermia affecting permeability of BBB. Methods A total of 72 healthy adult SpragueDawley rats were randomly divided into four groups,ie,sham operation group(SO group),TBI group,hyperthermia group(HT group)and mild hypothermia group(MH group).The rats were sacrificed at 24hours and the samples of injured brain tissues collected to detect the content of Evans blue(EB),activity of MMP-9 and expression of laminin. Results The content of EB and activity of MMP-9 in HT group were more significantly increased compared with those in TBI group(P<0.05),while the expression of laminin in HT group Was more significantly decreased than that in TBI group(P<0.05).However,the content of EB and activity of MMP-9 in MH group were significantly lower than those in TBI group(P<0.05),while the expression of laminin in MH group was significantly higher than that in TBI group(P<0.05). Conclusions The high activity of MMP-9 can.degrade laminin and increase permeability of BBB after TBI.After TBI,hyperthermia can increase permeability of BBB while mild hypothermia does on the contrary,which may be related to their different roles in changing activity of MMP-9.

ABSTRACT

Objective To investigate the clinical effects of tendon sheath laceration by syringe needle under the guidance of ultrasonography in the treatment of stenosing tenosynovitis. Methods A retrospective comparison was made on the curative efficacy in 78 patients with stenosing tenosynovitis of the hand. The patients were divided into two groups: the treatment group (n=39) was given tendon sheath laceration by 20-gauge syringe needle under the guidance of 10 MHz ultrasonography, while the control group ( n= 39) received the conventional puncture and laceration without the ultrasound guidance. Results At the fourth postoperative week, complete pain relief was achieved in 32 patients in the treatment group (82.1%) and in 20 patients in the control group (51.3%), respectively, with a significant difference ( ? 2 =8.34, P =0.004). The visual analogue score (VAS) was 8.9~3.5 (mean, 6.7?2.0) lower than before operation in the treatment group and 8.5~0.5 (mean, 5.2?2.9) in the control group. The pain relief in the treatment group is significantly superior to that in the control group ( t =2.659, P =0.010). Conclusions Ultrasound-guided laceration of the stenotic tendon sheath by syringe needle is a simple, minimally invasive and effective option for the management of stenosing tenosynovitis of the hand.

ABSTRACT

Objective:To clone human interleukin-33(hIL-33)and express it in E.coli efficiently.Methods:The primers were synthesized according to the hlL-33 cDNA sequence in GeneBank.The hIL-33 was amplified by RT-PCR from human fibroblast cell line(L929),the PCR product was inserted into pUC19 vector.The IL-33 cDNA confirmed by sequencing was inserted into expressing vector PQE30 and expressed in E.coli M15 strain.IL-33 protein expression was induced by IPTG and purified by Ni-NTA affinity chromatography.The recombinant IL-33 was identified by Immunoblot and its biological activity was analyzed.Results:DNA sequencing confirmed that the cloned cDNA was identical to the published sequence of hIL-33.The recombinant plasmid PQE/hIL-33 was transformed into M15.An expected 18KD protein of hIL-33 found mainly in the induced host strains about 25% of total bacteria lysis by SDS-PAGE and coomassie blue staining.The 18 KD protein could be recognized by anti-IL33 antibody in western blot.The recombinant protein was purified to more than 95% of total protein and induces the production of IL-4 and IL-5 in human peripheral blood mononuclear cells.Conclusion:We have successfully expressed hIL-33 protein in E.coli and the expressed product has IL-33 specific bioactivity.
